Effect of butanolic extract of Pueraria tuberosa DC. on the oestrous cycle of adult rats

1. The effect of butanolic extract of Pueraria tuberosa DC has been studied on the duration of oestrous cycle in adult cyclic rats. 2. The extract at 150 mg/kg dose induced cornified phase at the initial days of treatment which persisted till the last day of treatment. 3. Induction and persistance of cornification in the oestrous cycle due to the treatment of butanolic extract has been accounted for its estrogenic nature.

Effect of chronic treatment of estrogen and progesterone on histamine receptor sensitivity in guinea-pig uterus

1. In the present investigation the influence of estrogen and progesterone on the histamine induced responses in guinea pig uterus was studied. 2. The responses to histamine were significantly decreased in preparations pretreated with estrogen (pD2=4.75) or progesterone (pD2=5.27). 3. Blockade of responses to histamine by mepyramine after estrogen (pA2=8.65) and progesterone pretreatment (pA2=9.1 1) were significantly less than the control. 4. Dose dependant contractile responses of guinea-pig uterus were obtained with 2 - (2 pyridyl) ethylamine (pO2=5.95) which decreased after estrogen pretreatment (pD2=4 .35) or progesterone pretreatment (pD2=4.87). 5. The data suggest a decrease in H1 receptor sensitivity in guinea pig uterus after estrogen or progesterone pretreatment.

Local anaesthetic activity of 9-alkylamino-1,2,3,4,-tetrahydroacridines: structure-activity relationship study

1. A series of 9-alkylamino-1, 2, 3, 4-tetrahydroacridines and analogues were synthesized and tested for local ansesthetic activity in rabbits and guinea pigs. 2. 9-n-Heptylamino-1, 2, 3, 4-tetrahydroacridines showed the maximum activity and were more potent than lignocaine. 3. The local anaesthetic activity seems to be partly influenced by the lipophilicity of the compounds.

Effect of thiophanate methyl on central nervous system

1. Thiophanate methyl decreased spontaneous motor activity and body temperature. 2. It had no effect on pentobarbitone sleeping time, forced coordinated motor activity, conditioned avoidence response,convulsions induced by electrical stimulation or by pentylenetetrazol and did not exhibit local anaesthetic activity, 3. It had a significant analgesic effect.

1. The per se reIaxant effect of dopamine and its different analogues, with different substituents on nitrogen atom and alpha carbon atom, was studied on the isolated guinea pig tracheal chain. Isoprenaline and ritodrine were used as standards for comparison. 2. All the dopamine analogues, like isoprenaline and ritodrine, produced dose-dependent relaxation.The relaxant effect produced by alpha-methyl-isopropyl- dopamine and dobutamine was quite comparable to ritodrine but was less than that of isoprenaline.The activity of phenyl-secondary-butyl-dopamine was some what less than that of ritodrine.

1. Autonomic over activity is known to precede overt hypertension, and with intervention at this stage it could be possible to delay and minimise further elevation in blood pressure. 2. In the present study propranolol 20 mg/day has been tried in first order relatives of hypertensives who showed hyperresponsiveness in manoevures like cold pressor test and Valsalva manoevure which involve reflex sympathetic activity. Propranolol was effective in reducing the exaggerated response without causing overt cardiovascular effects.
