Anticonvulsant activity of dimethindene maleate (Foristal – Ciba)

A number of antihistaminics have occasionally been tried in the treatment of epilepsy and related disorders (Douglas, 1965). Vimal chandra, Bapat and Dhawan (1969) hare reported the effectiveness of a large number of antihistaminics against Supra-maximal Electroshock Serzure Pattern test (SMES test). In our previous study we had also indicated that the anticonvulsant activity is unrelated to antihistaminic activity of this class of compounds (Vimal Chandra, Bapat and Dhawan, 1969; 1970). In the present study the anticonvulsant action of dimethindene against SMES test, Metrazole seizure threshold test (MST) and 5-HT induced convulsions was determined and compared with that of diphenylhydantoin, diphenhydramine and cyproheptadine. A preliminary report of the part of this work has already been published (Vimal Chandra. Bapat. Ansari and Janhari, 1970).

I311 uptake was measured 4 and 24 hours after the injection of radioactive iodine in rats subjected to restraint stress for 18 hours after pretreatment with chlorpromazine (2 and 10 mg/kg) alone and in combination with restraint stress. In all these animals 1131 uptake hours, the reduction being 'more was reduced both after 4 and 24 marked after 4 hours.
