Seventeen antihistamines were studied far their effect on acetylcholine (Ach) Induced contraction of skeletal muscle (Skm) under Gaddum«SQ»s superfusion technique. All drugs exhibit some degree of cuare like action In 0.1 to 10 mcg dose except in pheniramine, mabhydroline and metiamide where there was a regression of curarimimetic effect with increasing doses. During recovery from drug effect the tissue showed an increased sensitivity to the action of Ach. In some instances this potentiation was immediate. In high doses (100 mcg and above). some of the antihistamines produced contracture of Skm which was not blocked by curare.
